The Potential of the Biotin-Avidin System (BAS) in Immunoassays

What is the Biotin-Avidin—System(BAS)?

The Biotin-Avidin System (BAS) is a widely used signal amplification system in immunoassays. At its core, Avidin, a common glycoprotein, consists of four identical subunits, each equipped with a biotin-binding site, making it exhibit a strong affinity for biotin.


Streptavidin, composed of four identical peptide chains, allows one Streptavidin molecule to bind to four biotin molecules. This binding property leads to an amplification effect, often employed as an intermediary in immunoanalyses.


One of the most prevalent applications involves immobilizing Avidin on the surface of magnetic beads while labeling antibodies with biotin. The high affinity between biotin and Avidin allows antibodies to be indirectly adsorbed onto the solid phase through this interaction.
Streptavidin molecules can also aggregate through non-covalent interactions, forming dimers, tetramers, and large aggregates.

As a result, the Avidin-biotin system can significantly amplify the signal response in immunoassays.
